Let's start with the basics.

Klotho is primarily expressed in the kidney, parathyroid and choroid plexus.

At the renal level, it is the co-receptor for Fibroblast Growth Factor 23 (FGF23).

Interaction with FGFR1c (Fibroblast Growth Factor Receptor 1c) leads to the formation of a complex that dimerizes with a second Klotho-FGF23-FGFR1c complex thanks to the help of Heparan Sulfate (HS).

Through phosphorylation, the FRS2-alpha --> Ras --> MAPK cascade is activated.
